Abstract: Modern drug discovery is an iterative, multi-disciplinary research endeavor that requires perseverance and commitment to successfully deliver new medicines to patients in need.  An overview of the drug discovery process, from disease hypothesis and initial ligand discovery through optimization and the identification of molecules with the right combination of properties to proceed through clinical evaluation will be presented. The trials, travails, and triumphs of the drug discovery process will be highlighted in a case study of Eli Lilly and Company’s BACE Inhibitor program, a promising drug target under investigation for the treatment of Alzheimer’s disease.
